PCRopsis Cell &
Bacterial Lysate in Urine

The PCRopsis Cell removes PCR inhibitors found in lysates from a mixture of bacterial and human cells to allow for the amplification of a bacteria-specific DNA fragment that would otherwise be undetectable.

A DNA fragment encompassing part of the bacterial 16S rRNA gene can only be amplified when the bacterial lysate is pre-incubated with PCRopsis Cell. Gene amplification was observed for both E. coli (gram negative) and S. aureus (gram positive) bacteria at ~100 and ~1000 cfu / sample. The urine sample contains human epithelial cells and therefore the lysate is a mixture of human and bacterial components. This result demonstrates that pre-incubation of lysates, containing a mixture of human and bacterial components, with PCRopsis Cell results in the expected amplification of a bacterial DNA target without further processing.

Note: PCR Study used PCRopsis 5x PCR Master Mix (cat. no. PMM001)
